Operational taxonomic units (OTUs): diversity units of 16S- or 18S-rDNA sequence sharing identity above a threshold; unless specified, usually 97%.
the 16S rRNA sequence reads can be clustered into OTUs at a 97% similarity level. These OTUs can be used to approximate the taxonomic rank species, although they do not precisely represent bacterial species. Each OTU is characterized by a representative DNA sequence and can be assigned a taxonomic lineage by comparing it with a known bacterial 16S rRNA database. We can further aggregate OTUs from the same genus and analyze the abundances at the genus level, which is more robust to sequencing error and can significantly reduce the number of variables in downstream analyses. Similarly, using the known database, OTUs can be assigned to families, orders, classes, or phyla
